Application of a Double Mannich Reaction Using Bis(aminol) Ethers in the Synthesis of AE Ring Analogues of Methyl Lycaconitine. — Double Mannich reaction
of preformed bis(aminol) ethers (I) with cyclic β-keto esters (II) and (V) represents an
efficient method for the synthesis of azabicyclic N-substituted ring systems (III) and
(VI), respectively. The reaction proceeds under mild conditions and provides the products in significantly improved yields as compared to conventional aminomethylation
protocols. The double Mannich reaction products (IIIe) and (VIe) are successfully applied in the synthesis of AE ring analogues of methyl lycaconitine [cf. (IV)]. —
