General and Efficient Indole Syntheses Based on Catalytic Amination Reactions.
The transformations proceed either via a palladium- and/or copper-catalyzed ami24- 111 —
nation reaction and are applicable to alkyl- and aryl-substituted alkynes and amines.
The copper-catalyzed procedure is more efficient for the bromo-substituted arenes. The
method allows the use of mild bases such as potassium phosphate or KOtBu and the
introduction of different protecting groups at nitrogen of the indole moiety. A one-pot
indole synthesis starting from ortho-chloroiodobenzene (IV) is viable using a single
catalytic system consisting of Pd(OAc)2/CuI/JDP and cesium carbonate affording (IIIa)
